I have a young weeping cherry tree that has developed a large shoot near the base of the tree. It's about 12" long and half as thick as a pencil. Is there any way I can propagate this shoot?

First, if you have a weeping cherry it is probably a "topgraft" with a regular cherry base/standard and grafted top that weeps. If so, then the sucker coming from the lower stem or root system is NOT a weeping cherry. I honestly don't know if you can do a softwood cutting that would actually take from this, but my thought is that you probably won't get the plant that you wanted - a weeper?
